DFHBI-1T IMAGES
A. Live-cell imaging of a COS7 cell expressing CGG60-Spinach2TM in the presence of either 20 µM DFHBI or DFHBI-1T. Images of the same cell were acquired using a 100 msec exposure with a GFP filter set. B. Quantification of average DFHBI-1T brightness of 10 cells expressing CGG60-SpinachTM normalized to the brightness of DFHBI.

Lucerna——活體胞內(nèi)RNA示蹤
綠色熒光蛋白的發(fā)現(xiàn)及其應(yīng)用是生命科學(xué)研究*一個(gè)重要的里程碑,使得細(xì)胞內(nèi)不可見(jiàn)的重要蛋白無(wú)所遁形,進(jìn)而明確其功能與調(diào)控機(jī)制。相較于蛋白質(zhì),近年在細(xì)胞內(nèi)還揭示了一大類重要的分子——非編碼RNA,這類特殊的RNA不編碼蛋白,但是其數(shù)目遠(yuǎn)較編碼RNA(mRNAs)多,包括microRNAs, Piwi-interacting RNAs, termini-associated RNAs等。目前對(duì)這一類RNA的研究方興未艾,因技術(shù)上缺乏關(guān)鍵的研究工具對(duì)其細(xì)胞內(nèi)定位于功能了解知之甚少。
胞內(nèi)RNA成像技術(shù)可能是非編碼RNA研究領(lǐng)域取得突破的關(guān)鍵,與綠色熒光蛋白相似,在目的RNA上標(biāo)記熒光分子可以使RNA在熒光顯微鏡下呈現(xiàn),明確其胞內(nèi)定位,進(jìn)而通過(guò)分子互作或藥理學(xué)等技術(shù)了解其功能。然而目前的常規(guī)RNA成像技術(shù)難以在活細(xì)胞上應(yīng)用,并且有效性存在很大問(wèn)題。針對(duì)這一難題,來(lái)自威斯康星大學(xué)的研究人員成立了Lucerna公司,并研發(fā)了一種*的基因編碼系統(tǒng)用于活細(xì)胞內(nèi)RNA的熒光標(biāo)記。該系統(tǒng)利用一種可與綠色熒光蛋白媲美的RNA原件——Spinach(一種RNA與源于綠色熒光蛋白的熒光小分子結(jié)合體,可與目的RNA結(jié)合并發(fā)射熒光)使得在活細(xì)胞中實(shí)時(shí)成像目的RNA的動(dòng)態(tài)機(jī)制。利用該系統(tǒng),研究人員可望在RNA胞內(nèi)轉(zhuǎn)運(yùn),降解、毒性RNA聚焦以及其他RNA調(diào)控機(jī)制的研究中取得突破性進(jìn)展。
